Cette force provient de filaments contractiles appel\u00e9s myofibrilles, tandis que l\u2019\u00e9nergie est fournie par les mitochondries. Ensemble, ils repr\u00e9sentent la majeure partie du contenu musculaire, permettant des contractions puissantes et soutenues. Le bon fonctionnement des muscles repose sur l\u2019organisation pr\u00e9cise des myofibrilles et des mitochondries dans un espace intracellulaire dense, sp\u00e9cifique \u00e0 chaque type de muscle. La mani\u00e8re dont cette organisation se met en place au cours du d\u00e9veloppement restait jusqu\u2019ici mal comprise. En utilisant la g\u00e9n\u00e9tique de la drosophile et l\u2019imagerie en direct, J\u00e9r\u00f4me Avellaneda a d\u00e9couvert que les microtubules coordonnent le transport des mitochondries avec la myofibrillogen\u00e8se.<br \/>Dans les muscles de vol de drosophile, les myofibrilles fibrillaires s\u2019alignent le long de l\u2019axe de contraction, avec des mitochondries intercal\u00e9es entre elles. Au d\u00e9but du d\u00e9veloppement, les microtubules forment de vastes r\u00e9seaux associ\u00e9s aux filaments d\u2019actine, qui s\u2019assemblent ensuite en myofibrilles. L\u2019imagerie en direct r\u00e9v\u00e8le que les mitochondries sont tr\u00e8s mobiles, se d\u00e9pla\u00e7ant le long des c\u00e2bles de microtubules et s\u2019intercalant rapidement entre les myofibrilles. Les muscles d\u00e9pourvus de moteurs de microtubules de type kin\u00e9sine pr\u00e9sentent des mitochondries regroup\u00e9es qui ne parviennent pas \u00e0 s\u2019intercaler, entra\u00eenant une architecture myofibrillaire d\u00e9fectueuse. La destruction pr\u00e9coce des microtubules perturbe l\u2019alignement des myofibrilles. Ces r\u00e9sultats identifient les microtubules comme des organisateurs cl\u00e9s des myofibrilles et des mitochondries au cours du d\u00e9veloppement musculaire. Une collaboration avec Edgar Gomes montre que ce r\u00f4le est conserv\u00e9 dans les<br \/>myofibres de souris en culture.
